Pharmacodynamic properties

Ketoprofen is a non-steroidal anti-inflammatory drug. In addition to its anti-inflammatory effect,  also exhibits antipyretic and analgesic effects. The pharmacological mechanism of action of ketoprofen is based on the inhibition of cyclo-oxygenase and lipoxygenase. Ketoprofen also prevents the formation of bradykinin and stabilizes the cell membranes of lysosomes, which prevents the release of lysosomal enzymes that mediate tissue destruction.

Pharmacokinetic properties

Ketoprofen is rapidly absorbed after intramuscular administration. It reaches its maximum plasma concentration in 30-60 minutes. Absolute bioavailability after intramuscular administration in cattle and swine is 90-100%, in horse 70%. The volume of distribution and excretion is approximately 0.17 L/kg and 0.3 L/kg, respectively. It shows linear kinetics. (So linear kinetics prevails, there is a linear relationship between the dose administered and the volume of distribution and excretion).

After intramuscular administration, the plasma half-life is 2-3 hours and is metabolized by secondary reduction to alcohol. It is thrown quickly. Mainly, 80% of the administered dose is excreted in the urine within 12 hours. The reduced ketoprofen metabolite is more abundant in cattle and the glucuronidated conjugate in horses.

AREA OF USE / INDICATIONS

Horse

Diseases affecting the osteoarticular and musculoskeletal system associated with acute pain and inflammation:

- lameness of traumatic origin

- Arthritis

- Osteitis, knee swelling

- Tendinitis, bursitis

- Naviculitis

- Laminitis

- Myositis

Ketoprofen is also indicated for the symptomatic treatment of postoperative inflammation, colic and fever.

Diseases related to inflammation, pain or fever

- Respiratory diseases

- mastitis

- For osteoarticular and musculoskeletal disorders such as lameness, arthritis and for easy postpartum standing

- Wounds

Pig

Diseases related to inflammation, pain or fever

- Postpartum Dysgalaxia Syndrome/Mastitis Metritis Agalaxia (MMA) Syndrome

- Respiratory tract infections

- Symptomatic treatment of fever

For short-term relief of post-operative pain associated with minor tissue surgery such as castration in piglets

 

Here, ketoprofen should be combined with appropriate antimicrobial therapy.

USAGE AND DOSAGE

Horse: intravenous injection

Cattle: intravenous or intramuscular injection

Pig: intramuscular injection

Horse:

For 3-5 days,  once a day,  2.2 mg ketoprofen body weight/day intravenously ie,

1 ml for every 45 kg of body weight

One injection is enough to treat colic. A reassessment of the patient's clinical status is required for the second administration of ketoprofen. See section 4.5, Special precautions for use.

Cattle:

3 mg ketoprofen body weight/day intravenously or deep intramuscularly, i.e. 3 ml per 100 kg body weight, once a day for 3 days.

Pig:

3 mg ketoprofen body weight/day as a single deep intramuscular injection, i.e. 3 ml per 100 kg body weight (=0.03 ml/kg).

To reduce post-operative pain, the product should be injected 10-30 minutes before surgery. Particular attention should be paid to the selection of the dosing device  (ie, low-dose syringe)  and accurate measurement of body weight in order to deliver the appropriate dose.

Do not use in case of hypersensitivity to the active substance or any excipient. Do not use in animals suffering from gastrointestinal lesions, hemorrhagic diathesis (weakness due to bleeding), weakened liver, kidney and heart function. It should not be used simultaneously with other NSAIDs, and if an NSAID has been administered, apply the product after 21 hours.
